چکیده
Asiatic citrus canker, one of the most important diseases citrus, is caused by Xanthomonas citri pv. citri. It has high economic impact and can spread easily, disease difficult to manage; it a quarantine organism in many citrus-producing countries. X. been separated into three subpathotypes (A, A * , w ) that differ host range geographical distribution, thus creating need differentiate for surveillance management. Availability useful diagnostic tools cornerstone successful surveillance, quarantine, eradication measures. In this study, multiplex conventional PCR (cPCR) assay was developed detection subpathotype determination Assay specificity assessed four different labs on total 146 58 other isolates. The demonstrated analytical sensitivity, specificity, selectivity. False negatives were observed with Lineage 2 strains, potential false positives bilvae. Combined simple extraction protocol, deployed successfully at Plant Protection Quarantine Pathogen Confirmatory Diagnostics Laboratory. This proven differentiating canker from symptomatic tissue. Development of a Multiplex RT-PCR Assay for Detection of the Causal Agents of Citrus Tristeza and Cachexia Diseases with Coamplification of Plant mRNA as an Internal Control
Background and Aims: Plant certification programs need reliable, fast, cheap and sensitive methods for detection of systemic pathogens with special interest in virus and viroid detection. Reverse transcriptase-polymerase chain reaction (RT-PCR) has been documented as an alternative assay for certification of plant propagating materials. The main object of the present study was the optimization ...
متن کاملSample Preparation for Secretome Analysis in A*-Type Strains of Xanthomonas citri subsp. citri, the Causal Agent of Asiatic Citrus Canker Disease
Xanthomonas citri subsp citri (Xcc) is responsible for one of the most destructive diseases of citrus fruits, Asiatic citrus canker. Based on the bacterial host range, the pathogenic forms, A, A*, and Aw have been identified. A*-type strains are not well characterized and they have been reported to differ from other A-type strains on the basis of biochemical and genetic characteristics. In this...
متن کاملQuantitative PCR method for diagnosis of citrus bacterial canker.
For diagnosis of citrus bacterial canker by PCR, an internal standard is employed to ensure the quality of the DNA extraction and that proper requisites exist for the amplification reaction. The ratio of PCR products from the internal standard and bacterial target is used to estimate the initial bacterial concentration in citrus tissues with lesions.
